The use of structured illumination (SI) can improve the performance of a fluorescence microscope through enhanced spatial resolution and the rejection of out of focus light.

We present an SI microscope in which excitation patterns are generated using a spatial light modulator.

We investigate the effect of light polarisation state on SI contrast and show how modification of both the illumination frequency and the image reconstruction algorithm can allow effective superresolution imaging in thick fluorescent samples.
